目的评价巢式聚合酶链反应(Nested--polymerase chain reaction,nested-PCR)在输入性卵形疟诊断和分型中的应用价值。方法采用巢式PCR方法回顾性检测湖北省2013年7月至2015年6月网报疟疾病例,比较疟疾快速诊断试剂盒(Rapid diagnostic tests,RDTs)、显微镜镜检和巢式PCR这三种方法对输入性疟疾的检测效果。结果巢式PCR共检出卵形疟24例,其中经典亚种10例,变异亚种14例;RDTs阳性13例,阴性11例;显微镜检15例诊断为卵形疟,8例为间日疟,1例为三日疟。卵形疟两亚种在RDTs诊断阳性率和镜检准确率差异无统计学意义(P>0.05)。结论巢式PCR在输入性卵形疟的诊断和分型上较RDTs和镜检法有显著优势。
Objective To evaluate the value of nested-PCR (Nested polymerase chain reaction) in the diagnosis and typing of imported ovipositive malaria. Methods Nested PCR was used to retrospectively detect netizens malaria cases from July 2013 to June 2015 in Hubei Province. Three rapid diagnostic tests (RDTs), microscopic examination and nested PCR were compared. Detection of imported malaria. Results There were 24 cases of oviparous malaria detected by nested PCR, including 10 cases of classical subspecies and 14 cases of variant subspecies; 13 cases of RDTs were positive and 11 cases were negative; 15 cases were diagnosed as oviparous malaria by microscopy and 8 cases were day Malaria, one case of Malaria on the 3rd. There was no significant difference in the diagnostic accuracy and the accuracy of microscopic examination between two ovales of oval-shaped malaria in RDTs (P> 0.05). Conclusion Nested PCR has significant advantages over RDTs and microscopy in the diagnosis and typing of imported oviparous malaria.
