目的研究肿瘤坏死因子(TNF)α多克隆抗体对新生大鼠缺氧缺血性脑损伤(HIBD)后大脑的保护效应。方法制备左脑HIBD的新生大鼠模型,于HIBD前后分别腹腔注射一定浓度的TNFα多克隆抗体,观察用药前后损伤大脑脑水含量及组织病理学变化。结果HIBD前后腹腔注射TNFα多克隆抗体可明显降低脑水含量(P<0.01),大脑水肿明显减轻,神经元死亡明显减少,但对其相应mRNA转录无影响。结论TNFα多克隆抗体可以拮抗TNFα的炎性作用,对HIBD后的大脑有保护效应
Objective To study the protective effects of polyclonal antibody against tumor necrosis factor α (TNF α) on the brain of neonatal rats after hypoxic-ischemic brain damage (HIBD). Methods The neonatal rat model of left HIBD was prepared. The polyclonal antibody against TNFα was injected intraperitoneally before and after HIBD. The changes of brain water content and histopathology were observed before and after treatment. Results The intraperitoneal injection of TNFα polyclonal antibody before and after HIBD significantly reduced the level of water in the brain (P <0.01). The cerebral edema was significantly reduced and the neuronal death was significantly reduced. However, it did not affect the mRNA transcription. Conclusions TNFα polyclonal antibody can antagonize the inflammatory effect of TNFα and protect the brain after HIBD
