目的 :通过观察脑胶质瘤中 p16mRNA及其蛋白、EGFR的表达 ,探讨其在脑胶质瘤发生、发展中的作用。 方法 :应用原位杂交技术和免疫组织化学方法在 38例人脑胶质瘤中检测 p16mRNA及其蛋白、EGFR的表达。以 7例正常组织作为对照。结果 :p16mRNA和 p16蛋白在Ⅲ~Ⅳ级、Ⅰ~Ⅱ级胶质瘤、正常组织中的阳性表达率分别为 8 3% ,16 7% ;5 7 7% ,5 7 7% ;71 4% ,85 7%。EGFR在Ⅲ~Ⅳ级、Ⅰ~Ⅱ级胶质瘤、正常组织中的阳性表达率分别为 83 3% ,38 5 % ,14 3%。结论 :p16基因缺失引起的 p16蛋白的表达降低、EGFR的过表达在脑胶质瘤的发生中起重要的作用。
OBJECTIVE: To observe the expression of p16mRNA and its protein, EGFR in glioma and to explore its role in the occurrence and development of glioma. Methods: In situ hybridization and immunohistochemistry were used to detect the expression of p16 mRNA, EGFR and EGFR in 38 human gliomas. Seven normal controls were used as controls. Results: The positive rates of p16 mRNA and p16 protein in grade Ⅲ ~ Ⅳ, grade Ⅰ ~ Ⅱ gliomas and normal tissues were 83%, 167%, 577%, 577%, 71 4% , 85 7%. The positive rates of EGFR in grade Ⅲ ~ Ⅳ, grade Ⅰ ~ Ⅱ gliomas and normal tissues were 83.3%, 38.5% and 143% respectively. Conclusion: The loss of p16 gene leads to the decrease of p16 protein expression. The overexpression of EGFR plays an important role in the pathogenesis of glioma.
