为探讨曲尼司特防治冠状动脉成形术后再狭窄及动脉粥样硬化的作用机制,以培养的大鼠血管平滑肌细胞为模型,应用氚标胸腺嘧啶脱氧核苷掺入实验及反转录-多聚酶链反应技术。观察致分裂原对血管平滑肌细胞增殖及C-myc基因表达的影响;并检测曲尼司特对致分裂原诱导的增殖及C-myc基因表达的抑制效应。结果显示,曲尼司特可明显抑制血小板源生长因子和血管紧张素Ⅱ诱导的DNA合成和C-myc基因表达。以上提示,曲尼司特能明显抑制血管平滑肌细胞增殖,其作用机制可能与抑制C-myc基因表达有关。
In order to explore the mechanism of tranilast in the prevention and treatment of restenosis and atherosclerosis after coronary angioplasty, the cultured rat vascular smooth muscle cells were used as a model, tritiated thymidine incorporation and reverse transcription - Polymerase chain reaction technology. To observe the effect of mitogen on the proliferation of vascular smooth muscle cells and the expression of C-myc gene, and to examine the inhibitory effect of tranilast on mitogen-induced proliferation and C-myc gene expression. The results showed that tranilast significantly inhibited platelet-derived growth factor and angiotensin II-induced DNA synthesis and C-myc gene expression. The above tips, tranilast can significantly inhibit vascular smooth muscle cell proliferation, its mechanism may be related to the inhibition of C-myc gene expression.
