用WatersPICOTAGTM系统分析测定了3种已知氨基酸序列的人重组肿瘤坏死因子的氨基酸组成,并测定了6批样品,对样品与对照品氨基酸组成的同质性及样品之间的一致性进行了统计分析比较,同时将所测得的蛋白质含量与用其它方法测定的结果进行了比较.结果表明,此法在检测基因工程产品的同一性方面是行之有效的,且具有快速、准确的特点,并能同时获得蛋白含量的测定结果.
The amino acid composition of human recombinant tumor necrosis factor (TNF) of three known amino acid sequences was analyzed by WatersPICOTAGTM system and six batches of samples were determined. The homogeneity of the amino acid composition of samples and reference samples and the consistency among the samples were calculated Analysis and comparison, while the measured protein content and other methods were compared with the results were compared.
