目的:研究在整体动物水平白细胞介素1(interleukin1,IL-1)对β淀粉样前体蛋白(βamyloidprecursorprotein,APP)基因表达的影响。方法:大鼠侧脑室注射;Northern杂交。结果:首次证实IL-1β在整体动物水平可以诱导大鼠脑组织APPmRNA表达的增加,而且这种诱导作用具有时间依赖性。结论:IL-1β在体内、外均可诱导神经组织内APP基因的表达增加,进而在阿尔茨海默病(Alzheimerdisease,AD)中与促进β淀粉样蛋白(βamyloidprotein,βAP)的沉积可能密切相关。
Objective: To investigate the effects of interleukin 1 (IL-1) on the gene expression of β-amyloid precursor protein (APP) in whole animal. Methods: Intracerebroventricular injection; Northern hybridization. Results: For the first time, it was confirmed that IL-1β could induce the increase of APPmRNA expression in rat brain at the level of whole animals, and the induction of this effect was time-dependent. CONCLUSION: IL-1β can induce the expression of APP gene in nerve tissue both in vitro and in vivo, which may be closely related to the promotion of the deposition of βamyloid protein (βAP) in Alzheimerdhosis (AD) .
