目的:探寻清肠化湿法对大鼠溃疡性结肠炎NF-κB、COX-2表达的影响,进而分析其可能的作用机制。方法:采用葡聚糖硫酸钠制备大鼠溃疡性结肠炎模型。实验设正常对照组、模型对照组、阳性药物组(柳氮磺吡啶组)、清肠化湿法组,每组8只。分别观察治疗后大鼠结肠组织NF-κB、COX-2的表达。结果:在正常结肠组织中NF-κB、COX-2呈弱表达,模型组结肠组织NF-κB、COX-2表达明显高于正常对照组,清肠化湿法组、SASP组结肠组织NF-κB、COX-2的表达明显低于模型组,清肠化湿法组、SASP组结肠组织NF-κB、COX-2的表达无明显差异。结论:清肠化湿方能抑制溃疡性结肠炎大鼠结肠组织NF-κB、COX-2的表达,可能是其疗效的机制之一。
Objective: To investigate the effect of Qingchang Huashi method on the expression of NF-κB and COX-2 in rat ulcerative colitis, and to analyze its possible mechanism. Methods: Dextran sodium sulfate was used to prepare ulcerative colitis model in rats. Experimental set up normal control group, model control group, positive drug group (sulfasalazine group), bowel dampness group, each group of eight. The expressions of NF-κB and COX-2 in the colon of the rats after treatment were observed. Results: The expression of NF-κB and COX-2 in normal colorectal tissues were weakly expressed. The expressions of NF-κB and COX-2 in colonic tissues of model group were significantly higher than those of normal control group. κB and COX-2 were significantly lower than those in the model group. There was no significant difference in the expression of NF-κB and COX-2 in the bowel between the Qingchanghuazhuo group and the SASP group. Conclusion: Qingchang Huashi prescription can inhibit the expression of NF-κB and COX-2 in the colon of rats with ulcerative colitis, which may be one of the mechanisms of its curative effect.
