目的探究同种异系异体小鼠皮肤移植后外周血中T淋巴细胞形态变化与移植排斥程度之间的关系,以期为诊断排斥反应提供一种新的无创检测方法。方法 203只清洁级健康雄性Balb/c小鼠,随机分为3组:空白对照组、自体移植组、异体移植组。分别于术后各时相点断尾取外周血在超显微真彩细胞分析仪下观察,寻找T淋巴细胞并录像记录其变化,得出相应T淋巴细胞形态变化指数(morphologic change index of T-lymphocyte,TMCI);同时处死小鼠,取移植皮片行组织病理检查,进行移植皮肤病理分级(skin pathology grade,SPG)。结果术后第1、2、3、6、7、8、9天异体移植组TMCI分别为0.086±0.01、0.090±0.01、0.082±0.01、0.113±0.02、0.156±0.07、0.119±0.01、0.118±0.04,较自体移植组(0.056±0.01、0.062±0.02、0.061±0.01、0.064±0.01、0.058±0.02、0.062±0.01、0.062±0.01)显著升高,P均﹤0.05;SPG与TMCI之间存在正相关,回归方程:SPG=24.615×TMCI-1.036。结论 TMCI可以代表T淋巴细胞的形态变化大小,且TMCI值越高对急性排斥反应的诊断越有意义。
Objective To investigate the relationship between the morphological changes of peripheral blood T lymphocytes and the degree of graft rejection after allogeneic allogeneic mouse skin transplantation in order to provide a new noninvasive detection method for the diagnosis of rejection. Methods Totally 203 healthy male Balb / c mice were randomly divided into 3 groups: blank control group, autograft group and allograft group. Peripheral blood samples were taken from the end of each time point after surgery and observed under ultra-microscopic true color cell analyzer. T lymphocytes were looked for and recorded. At the same time, the mice were sacrificed and the skin grafts were taken for histopathological examination and skin pathology grade (SPG) was performed. Results The TMCI of allogeneic transplantation group were 0.086 ± 0.01, 0.090 ± 0.01, 0.082 ± 0.01, 0.113 ± 0.02, 0.166 ± 0.07, 0.119 ± 0.01 and 0.118 ± 0.04, P <0.05 compared with autologous transplantation group (0.056 ± 0.01,0.062 ± 0.02,0.061 ± 0.01,0.064 ± 0.01,0.058 ± 0.02,0.062 ± 0.01,0.062 ± 0.01); there was a significant difference between SPG and TMCI Positive correlation, regression equation: SPG = 24.615 × TMCI-1.036. Conclusion TMCI can represent the size of T lymphocytes, and the higher the TMCI value, the more meaningful the diagnosis of acute rejection.
