目的 研究矽肺患者肺泡灌洗液中肺泡巨噬细胞 (AM )对人胚肺成纤维细胞 (FB)增殖的影响。方法 将AM分为加尘组 (S组 )和不加尘组 (O组 ) ,其中S组用 30 μg/ml二氧化硅粉尘刺激 ,培养 1 ,2 ,6 ,1 2 ,2 4 ,36h ,收集培养上清。用MTT法、生长曲线法和分裂指数法观察AM上清对FB的作用。结果 除O组 1h外 ,其余各组上清均能刺激FB增殖。从整体趋势上 ,S组促增殖作用强于O组。S组和O组的作用高峰分别出现在 6h和 1 2h。结论 矽肺患者AM培养上清的促增殖能力与AM的体外培养时间有关。AM体外培养过程可能是一个生长因子积累的过程 ,诸多细胞因子导致FB增殖。
Objective To study the effect of alveolar macrophages (AM) in bronchoalveolar lavage fluid on the proliferation of human embryo lung fibroblasts (FB). Methods AM was divided into dusting group (group S) and non-dusting group (group O). S group was stimulated with 30 μg / ml silica dust and cultured for 1, 2, 6, 12, 24, 36h , Collect the culture supernatant. The effects of AM supernatant on FB were observed by MTT assay, growth curve assay and split index method. Results In addition to O group 1h, the rest of the supernatant can stimulate FB proliferation. From the overall trend, the role of S group to promote proliferation stronger than the O group. The peak of S group and O group appeared at 6h and 12h respectively. Conclusion The proliferative capacity of AM supernatant in patients with silicosis is related to the in vitro culture time of AM. AM in vitro culture process may be a process of accumulation of growth factors, many cytokines lead to FB proliferation.
