目的了解厦门地区学龄前儿童细胞色素P450 3A酶(CYP3A)及分布活性特征。方法用氢化可的松(HC)非侵入性探针法,利用高效液相色谱(HPLC)测定尿液中6β-羟基氢化可的松(6β-OHC)、氢化可的松(HC)的浓度,以两者的浓度比定量表示CYP3A酶活性。结果 287名儿童酶活性呈左偏态分布,中位数为2.61,四分位数间距为3.97。结论不同性别间酶活性水平无显著性差别;随着年龄的升高,酶活性水平呈现降低的趋势(P<0.05),与年龄具有相关性;3岁组明显高于4岁组(P<0.05)。
Objective To understand the characteristics of cytochrome P450 3A enzyme (CYP3A) and its distribution activity in preschool children in Xiamen area. Methods The concentrations of 6β-hydroxy-hydrocortisone (6β-OHC) and hydrocortisone (HC) in urine were determined by high performance liquid chromatography (HPLC) using noninvasive probe method of hydrocortisone The CYP3A enzyme activity was quantified by the concentration ratio of both. Results The enzyme activity of 287 children showed a left-skewed distribution with a median of 2.61 and a quartile spacing of 3.97. Conclusion There was no significant difference in the levels of enzyme activity between different sexes. The enzyme activity level decreased with age (P <0.05), and was correlated with age. The 3-year-old group was significantly higher than the 4-year old group (P < 0.05).
