目的观察感染尖吻蝮蛇舌形虫小鼠体内特异性抗体和循环抗原动态变化。方法从感染尖吻蝮蛇舌形虫的五步蛇体内收集成虫,分离尖吻蝮蛇舌形虫的虫卵感染小鼠,从感染后1w、2w、3~22w收集小鼠血清,分别用ELISA法和dot-ELISA法观察不同时间小鼠体内尖吻蝮蛇舌形虫特异性抗体和循环抗原的动态变化。结果感染尖吻蝮蛇舌形虫的小鼠,其体内特异性抗体从第8w开始上升,12w达到高峰,第16w开始下降并一直维持同一水平。同时,对小鼠产生的特异性抗体进行分型,其最早出现为IgM,16w以后被IgG1所替代。感染尖吻蝮蛇舌形虫的小鼠血清用dot-ELISA法检测其循环抗原出现的时间为第1w,到第3w时循环抗原检出稀释度在1∶8~1∶128之间,到第8w,最高稀释度可达到1∶256,并一直维持,第11w以后逐渐下降。结论1.小鼠感染尖吻蝮蛇舌形虫虫卵后特异性抗体(Ab)在感染第8w开始上升,抗体最高滴度维持时间为第12~15w。2.感染尖吻蝮蛇舌形虫虫卵的小鼠其血清中产生的特异性抗体最早出现为IgM,以后被IgG1所替代。3.小鼠在感染尖吻蝮蛇舌形虫虫卵后的1w就可在其血清中检测到循环抗原(CAg),预测这段时间检测循环抗原具有早期诊断参考价值。
Objective To observe the dynamic changes of specific antibodies and circulating antigens in mice infected with Trichinella spiralis. Methods The adult worms were collected from five-step snake infected with Trionyx japonicus, and the mice were infected with the eggs of H. gondii. The sera of mice were collected from 1w, 2w and 3 ~ 22w after infection, ELISA and dot-ELISA method were used to observe the dynamic changes of specific antibodies and circulating antigens of H. gondii at different time points. Results In mice infected with Agkistrodon acutus, their specific antibodies increased from the 8th day to the 12th day and began to decline at the 16th day and kept the same level. At the same time, the specific antibodies produced by mice were typed, the earliest appeared as IgM, after 16w was replaced by IgG1. The serum of mice infected with Agkistrodon acutus was detected by Dot-ELISA for the first 1w of the circulating antigen, and the dilution of circulating antigen was detected between 1: 8 and 1: 128 at 3w, 8w, the highest dilution of up to 1:256, and has been maintained, gradually decreased after the 11w. The results showed that the specific antibody (Ab) of mouse infected with Agkistrodon acutus eggs increased from 8th week of infection, and the highest titer of antibody maintained for 12th to 15th week. 2. The specific antibodies produced in the serum of mice infected with Agkistrodon flacumolyticus eggs first appeared as IgM and later replaced by IgG1. In mice infected with Agkistrodon flaperinae eggs 1w after 1w can be detected in the serum of circulating antigen (CAg), predict the detection of circulating antigen during this period with early diagnosis of reference value.
