目的:建立高效毛细管电泳测定地榆中没食子酸含量的方法。方法:优化出毛细管电泳的最佳测定条件及参数:PDA检测器,检测波长为220 nm;电泳缓冲液体系为20 mmol·L-1的硼砂溶液(p H 9.0);运行电压为15 k V;选取苯甲酸钠为内标物定量。结果:没食子酸在0.200~6.000μg·ml-1范围内线性良好(r=0.999 6),LOD为0.050μg·ml-1,精密度RSD≤1.72%,地榆的平均加样回收率为98.78%(RSD=2.57%,n=3);测得地榆中没食子酸含量为0.31%。结论:该方法准确,可靠,重复性好,可用于地榆中没食子酸含量的测定。
Objective: To establish a method for the determination of gallic acid in Burnet by high performance capillary electrophoresis. Methods: The optimum conditions and parameters of capillary electrophoresis were optimized: PDA detector with a detection wavelength of 220 nm; electrophoresis buffer system was 20 mmol·L-1 borax solution (p H 9.0); the operating voltage was 15 kV ; Sodium benzoate as internal standard quantitative. Results: The linearity was good (r = 0.999 6), LOD was 0.050 μg · ml-1, RSD was less than or equal to 1.72%, and the average recoveries of gallic acid in the range of 0.200-6.000μg · ml-1 were 98.78 % (RSD = 2.57%, n = 3). The content of gallic acid in Burnet was 0.31%. Conclusion: The method is accurate, reliable and reproducible. It can be used for the determination of gallic acid in Burnet.
