目的 观察大鼠脑内星形胶质细胞对上唇皮肤伤害性刺激的反应及其与神经元的关系 .方法 将 Formalin注入大鼠一侧上唇皮下 ,成活 1,3,6 h后 ,用抗 FOS蛋白、抗酪氨酸羟化酶 (TH)和抗胶质原纤维酸性蛋白 (glial fibrillaryacidic protein,GFAP)的三重免疫组化方法显示反应神经元与星形胶质细胞的定位及其相互关系 .结果 1FOS阳性神经元和 GFAP阳性星形胶质细胞均特异地表达于与痛觉传递或镇痛相关的机能区 ;2三重免疫组化显示反应性神经元(FOS阳性 )与反应性星形胶质细胞 (GFAP阳性 )关系密切 ,形成三种复合体 .即 TH+ / FOS+ / GFAP+三标记复合体、TH+ / GFAP+ / FOS-二标记复合体和 FOS+ / GFAP+ / TH-二标记复合体 .3Formalin注射后 3h,复合体的数目达高峰 .结论 神经元 -星形胶质细胞复合体在口唇伤害性刺激信息处理过程中可能起着重要作用
Objective To observe the response of rat astrocytes to noxious stimulation of upper lip skin and its relationship with neurons.Methods Formalin was injected subcutaneously into the rat’s upper lip for 1, Triple immunostaining of proteins, anti-tyrosine hydroxylase (TH), and glial fibrillary acid protein (GFAP) revealed the location of reactive neurons and astrocytes and their correlation. Results Both 1FOS-positive neurons and GFAP-positive astrocytes were specifically expressed in functional areas associated with pain sensation or analgesia; 2T3 immunohistochemistry showed that reactive neurons (FOS positive) and reactive astrocytes (GFAP positive), and formed three complexes, namely TH + / FOS + / GFAP + triple labeling complex, TH + / GFAP + / FOS- two labeling complex and FOS + / GFAP + / TH- 3h, and the number of complex reached the peak.Conclusion Neuronal-astrocyte complex may play an important role in the process of lip noxious stimulation information processing
