目的:探讨参芎化瘀胶囊对血管性痴呆(vascular dementia,VD)大鼠海马CA1区p38丝裂原活化蛋白激酶(mitogen activated protein kinase,MAPK)表达的影响。方法:采用反复夹闭双侧颈总动脉同时ip硝普钠的方法制备血管性痴呆大鼠模型。SD大鼠随机分为:正常组、假手术组、痴呆模型组(模型组)、参芎化瘀胶囊治疗组。应用Morris水迷宫检测VD大鼠学习记忆能力,免疫组化染色检测海马CA1区p38MAPK表达的变化。结果:(1)与正常对照组及假手术组相比,模型组大鼠平均逃避潜伏期时间延长(P<0.01),跨越平台的次数及在原平台象限停留时间明显减少(P<0.01,P<0.01);与模型组比较,参芎化瘀胶囊治疗组随时间延长大鼠平均逃避潜伏时间缩短(P<0.05,P<0.01),穿越平台的次数及在原平台象限停留时间增多(P<0.05,P<0.05)。(2)与正常组及假手术组相比,模型组大鼠海马CA1区p38MAPK阳性细胞明显增多(P<0.01);与模型组比较,参芎化瘀胶囊治疗组大鼠海马CA1区p38MAPK阳性细胞减少(P<0.01)。结论:VD大鼠学习记忆成绩下降与p38MAPK表达增加有关,参芎化瘀胶囊可减少VD大鼠海马CA1区p38MAPK的表达,改善VD大鼠的学习记忆功能。
Objective: To investigate the effect of Shenqi Huayu Capsule on the expression of p38 mitogen-activated protein kinase (MAPK) in hippocampal CA1 region of vascular dementia (VD) rats. METHODS: A rat model of vascular dementia was prepared by repeatedly clamping both common carotid arteries with sodium nitroprusside. SD rats were randomly divided into normal group, sham operation group, dementia model group (model group), Shenqihuayu capsule treatment group. The learning and memory abilities of VD rats were detected by Morris water maze. The expression of p38MAPK in hippocampal CA1 region was detected by immunohistochemistry. Results: (1) Compared with the normal control group and the sham operation group, the average escape latency time of the model group rats was prolonged (P<0.01), the number of crossing platforms and the residence time in the original platform quadrant were significantly reduced (P<0.01, P< 0.01); compared with the model group, Shenqi Huayu capsule treatment group rats over time, the average escape latency shortened (P <0.05, P <0.01), the number of crossing the platform and the residence time in the original platform quadrant increased (P <0.05 P<0.05). (2) Compared with normal group and sham operation group, the expression of p38MAPK positive cells in hippocampal CA1 region was significantly increased in model group rats (P<0.01). Compared with model group, p38MAPK positive in hippocampal CA1 region of rats in Shenqihuayu capsule group Decrease in cells (P < 0.01). Conclusion: The decrease of learning and memory in VD rats is related to the increase of p38MAPK expression. Shenqi Huayu Capsule can reduce the expression of p38MAPK in hippocampal CA1 region of VD rats and improve the learning and memory function of VD rats.
