我们从人脑和肝组织中分离获得的神经节苷脂单组分GM_1和GM_3对兔红细胞膜钠泵活性影响进行了研究。GM_1的组成中比GM_3多一个半乳糖-N-乙酰半乳糖残基。实验表明:GM_1对兔红细胞膜钠泵有激活作用,而GM_3则为非竞争性抑制作用,V_m发生改变而K_m不变。提示外源性GM_1和GM_3的掺入,对钠泵的激活和抑制作用,可能通过诱导细胞膜微环境的改变,或直接结合于膜上的特殊位点,引起钠泵构象的改变而影响其活性。
We studied the effect of ganglioside GM_1 and GM3, which were isolated from human brain and liver tissue, on the activity of sodium pump in rabbit erythrocyte membrane. There is one more galactose-N-acetylgalactose residue in the composition of GM_1 than GM_3. Experiments show that: GM_1 has an activation effect on rabbit erythrocyte membrane sodium pump, while GM_3 is a non-competitive inhibition, V_m changes and K_m does not change. It is suggested that the incorporation of exogenous GM_1 and GM_3 and the activation and inhibition of sodium pump may affect the activity of the sodium pump by inducing changes in the cell membrane microenvironment or directly binding to specific sites on the membrane, causing changes in the sodium pump conformation. .
