目的:通过正交试验设计的方法,采用RP-HPLC法测定老鹳草酚酸类成分的含量并优选其提取工艺。方法:通过方法学考察,采用乙醇回流提取的方法,以酚酸类成分总含量为评价指标,以乙醇浓度、料液比、提取时间和提取次数为考察因素,通过正交试验筛选出最佳提取工艺。结果:最佳提取工艺为回流提取2 h、料液比1∶40、80%乙醇、提取2次。结论:优化后的提取方法节约物料,操作简单,不仅可以为工业大规模生产提供一定的实验数据,也为药材的进一步开发利用和质量评价提供理论依据。
OBJECTIVE: To determine the contents of phenolic acids in the genus Geranium by RP-HPLC and to optimize its extraction process by means of orthogonal design. Methods: By methodological study, the method of ethanol reflux extraction and the total content of phenolic acids were used as evaluation indexes. The ethanol concentration, the ratio of solid to liquid, the extraction time and the number of extraction were taken as the investigation factors, and the best Extraction process. Results: The best extraction process was reflux extraction 2 h, solid-liquid ratio 1:40, 80% ethanol, extracted twice. Conclusion: The optimized extraction method can save materials and has simple operation, which not only can provide some experimental data for large-scale industrial production, but also provide theoretical basis for further development, utilization and quality evaluation of medicinal materials.
