目的通过检测槟榔碱对胎鼠外周血细胞DNA损伤的程度和观察胎鼠生长发育情况,探讨槟榔碱对小鼠胚胎毒性的影响。方法使用槟榔碱给雌鼠灌胃6-8周后,与雄鼠合笼,孕16d后各组随机取孕鼠6只,剖腹取胎鼠,检测各组胎鼠的外周血细胞DNA损伤情况,并进行分析。孕19d后处死各组孕鼠,检测胎鼠的生长发育情况以及各组胎鼠的畸形发生率。结果正常的对照组胎鼠外周血细胞DNA出现典型的原型,而实验组的胎鼠外周血细胞DNA出现特征性的彗星形态。于孕19d处死各组动物,发现实验组动物胎鼠的生长发育指标包括身长、体重、尾长小于对照组,畸形发生率高于对照组。结论槟榔碱能对胎鼠外周血DNA造成一定程度的损伤,并对胎鼠各项生长发育指标均有影响,推测槟榔碱对胚胎产生一定毒性影响。
OBJECTIVE: To investigate the effects of arecoline on DNA damage in fetal rat peripheral blood cells and to observe the growth and development of fetal rat embryos and to explore the effects of arecoline on mouse embryotoxicity. Methods The rats were intragastrically administered with arecoline for 6-8 weeks and then caged with male rats. Sixteen pregnant rats were randomly selected from each group after pregnancy for 16 days. Fetal rats were taken by cesarean section to detect the DNA damage of peripheral blood cells in each group. And analyze. Pregnant rats were sacrificed on day 19 after pregnancy, and the growth and development of fetal rats and the incidence of deformity in each group were observed. Results Normal control group of fetal rat peripheral blood cell DNA typical prototype, and the experimental group of fetal rat peripheral blood cell DNA appeared characteristic comet morphology. Animals were sacrificed at 19d and found that the growth and development of the test animals, including body length, body weight and tail length, were smaller than those of the control group. The incidence of deformity was higher than that of the control group. Conclusion Arecoline can damage the fetal rat peripheral blood DNA to a certain degree and affect the growth and development of fetal rats. It is presumed that arecoline can cause some toxic effects on embryos.
