利用柯斯质粒载体在大肠杆菌中克隆和分析链霉菌基因簇的方法和技术已有很大发展,已构建了不少链霉菌-大肠杆菌双功能柯斯载体.利用这种载体固然可以把链霉菌的基因文库构建在大肠杆菌中并可方便地进行克隆片段的结构分析,但要通过转化把克隆的大片段DNA 转回到链霉菌中来进行功能分析常常遇到不少障碍,最近发展起来的质粒在大肠杆菌和链霉菌之间进行属间接合转移的方法,则解决了外源DNA导入许多链霉菌菌种的疑难问题,使得DNA的克隆和分析工作能够方便地在大肠杆菌中完成
