目的本研究旨在探讨腹腔注射乳化异氟醚对大鼠气道上皮细胞中β防御素BD-1、BD-2以及促炎症因子TNF-α、IL-1β、IL-8 mRNA表达水平的影响。方法健康清洁级SD大鼠36只随机分成6组,分别为A~F组。A组经腹腔注射生理盐水,B组经腹腔注射30%脂肪乳,C~F组分别经腹腔注射0.25、0.5、1.0、1.5倍ED50量8%乳化异氟醚。各组均于注药后1 h处死大鼠,取其气管黏膜组织。从每个组收获的组织中检测β防御素BD-1、BD-2和促炎症因子TNF-α、IL-1β、IL-8的mRNA表达水平。结果在腹腔注射乳化异氟醚的大鼠气管上皮组织中除BD-1和IL-8 mRNA无变化外,BD-2和促炎症因子TNF-α,IL-1βmRNA表达水平均增加,且随乳化异氟醚用量的增加而增强。结论本实验说明腹腔注射乳化异氟醚能在转录水平上调节大鼠气道上皮组织β防御素及一些促炎症因子mRNA的表达水平,且乳化异氟醚以剂量依赖的方式显著增加大鼠气道上皮细胞的β防御素BD-2以及这些促炎症因子mRNA的表达水平。
Objective To investigate the effects of intraperitoneal injection of isoflurane on the expression of β-defensin BD-1, BD-2 and the proinflammatory cytokines TNF-α, IL-1β and IL-8 mRNA in rat airway epithelial cells . Methods Thirty-six healthy and clean SD rats were randomly divided into 6 groups: A ~ F group. Rats in group A were injected intraperitoneally with normal saline. Group B received intraperitoneal injection of 30% fat emulsion. Groups C to F were injected intraperitoneally with 0.25, 0.5, 1.0, 1.5 times ED50 and 8% emulsified isoflurane respectively. Rats in each group were sacrificed at 1 h after injection, and their tracheal mucosa was taken. The levels of mRNA expression of beta defensin BD-1, BD-2 and the proinflammatory cytokines TNF-α, IL-1β, IL-8 were measured from the tissues harvested from each group. Results BD-2 and proinflammatory cytokines TNF-α and IL-1βmRNA expression increased in rat tracheal epithelial cells injected intraperitoneally with isoflurane, except BD-1 and IL-8 mRNA, Isoflurane dosage increased. Conclusion This experiment shows that intraperitoneal injection of isoflurane can regulate the mRNA expression of β-defensin and some proinflammatory cytokines in the airway epithelium of rats at the transcriptional level, and emulsified isoflurane significantly increases the expression of rat gas in a dose-dependent manner Beta defensin BD-2 of the epithelial cells and the expression levels of these proinflammatory cytokines.
