目的研究藤黄酸对人胃癌BGC-803细胞增殖和转移相关能力的影响及其机制。方法 MTT、Hoechst染色法检测藤黄酸对BGC-803细胞的增殖和凋亡的影响;侵袭小室模型等方法检测藤黄酸对BGC-803细胞侵袭、黏附、迁移的影响;运用RT-PCR、Western-blotting法分析藤黄酸对胃癌细胞凋亡和转移相关基因bcl-2、bax、细胞黏附分子-1(ICAM-1)mRNA和蛋白表达的影响。结果 藤黄酸抑制BGC-803细胞的生长,其抑制率与药物浓度及作用时间呈依赖关系。低浓度的藤黄酸处理BGC-803细胞后,可明显降低胃癌细胞的黏附、迁移和侵袭能力。随着藤黄酸浓度的增大,bcl-2I、CAM-1 mRNA和蛋白表达均下调,bax mRNA和蛋白表达上调。结论藤黄酸对胃癌细胞BGC-803有明显的生长抑制作用,具有显著的促肿瘤细胞凋亡作用和抑制肿瘤细胞转移相关性质,其机制可能与下调bcl-2I、CAM-1及上调bax的表达有关。
Objective To study the effects of gambogic acid on the proliferation and metastasis of human gastric cancer cell line BGC-803 and its mechanism. The effects of gambogic acid on the proliferation and apoptosis of BGC-803 cells were assayed by MTT and Hoechst staining. The invasion chamber cell model was used to detect the invasion, adhesion and migration of BGC-803 cells. RT-PCR, The effects of gambogic acid on the mRNA and protein expression of apoptosis-related and metastasis-related genes bcl-2, bax and ICAM-1 in gastric cancer cells were analyzed by Western-blotting. Results Gambogic acid could inhibit the growth of BGC-803 cells. The inhibitory rate was dependent on the drug concentration and time. Treatment of BGC-803 cells with low concentrations of gambogic acid could significantly reduce the adhesion, migration and invasion ability of gastric cancer cells. With the increase of concentration of gambogic acid, the expressions of bcl-2I, CAM-1 mRNA and protein were down-regulated and the expression of bax mRNA and protein were up-regulated. Conclusion Gambogic acid has obvious growth inhibitory effect on BGC-803 gastric cancer cells, and has the obvious function of promoting apoptosis of tumor cells and inhibiting the metastasis of tumor cells. The mechanism may relate to down-regulation of bcl-2I, CAM-1 and up-regulation of bax Expression related.
