目的:观察天麻乙酸乙酯提取物对大脑中动脉栓塞模型大鼠神经突触可塑性的影响。方法:将动物随机分为3组,即假手术组、模型组、天麻乙酸乙酯提取物组(以下简称乙提组,剂量按生药量计为7.29 g·kg-1)。预防性连续灌胃给予药物7 d,末次给药0.5 h后复制大鼠大脑中动脉栓塞模型,然后再进行治疗性连续灌胃给药7 d,统计模型复制后7 d时的死亡率、各组大鼠在规定时间点进行神经行为学评定及脑组织取材检测GAP-43的表达。结果:与假手术组比较,模型组大鼠死亡率和神经病学评分明显升高、肌力评分和GAP-43表达明显降低;与模型组比较,乙提组大鼠术后第7天死亡率和神经病学评分明显降低、肌力评分和GAP-43表达明显升高,差异有统计学意义(P<0.05,P<0.01)。结论:天麻乙酸乙酯提取物对大脑中动脉栓塞模型大鼠神经突触可塑性有一定的改善作用。
Objective: To observe the effect of ethyl acetate extract of Gastrodia elata Blume on the synaptic plasticity of middle cerebral artery occlusion model rats. Methods: The animals were randomly divided into three groups: sham operation group, model group, ethyl acetate extract of Gastrodia elata Blume (hereinafter referred to as B group, the dose of crude drug was 7.29 g · kg-1). The drug was administered prophylactically continuously for 7 days. The model of middle cerebral artery occlusion was reproduced after 0.5 h of the last administration. The rats were then given a therapeutic continuous gavage for 7 days. The mortality rate at 7 days after the model was replicated Group rats at the prescribed time point neurobehavioral assessment and brain tissue samples were taken to detect the expression of GAP-43. Results: Compared with the sham operation group, the mortality and neurological score of the model group were significantly increased, and the muscle strength score and the expression of GAP-43 were significantly decreased. Conclusion: The extract of Gastrodia elata Blume can improve the synaptic plasticity of middle cerebral artery occlusion model rats.
