目的 观察TGF β和胶原酶、TIMP 1mRNA在增生性瘢痕中的表达 ,探讨TGF β对MMP 1和TIMP 1表达的影响以及它们在HTS形成中的作用。方法 采用原位杂交和图像分析技术 ,进行定性和定量分析。结果 增生性瘢痕组织中TGF β和TIMP 1表达较正常皮肤明显增高 ,平均积分光密度分别增加了 8倍和 7倍 ,也较胶原酶的表达明显增高 ,相差非常显著 ,TGF β表达与TIMP 1表达呈高度正相关。结论 TGF β可能促进TIMP 1表达 ,抑制胶原酶活性 ,阻止胶原降解 ,是HTS形成的重要原因。
Objective To observe the expression of TGFβ and collagenase and TIMP-1 mRNA in hypertrophic scars and to explore the effect of TGFβ on the expression of MMP-1 and TIMP-1 and their roles in the formation of HTS. Methods In situ hybridization and image analysis techniques were used for qualitative and quantitative analysis. Results The expression of TGF β and TIMP 1 in hypertrophic scar tissue was significantly higher than that in normal skin. The average integrated optical density increased 8-fold and 7-fold, respectively. Compared with collagenase, the expression of TGF-β and TIMP-1 were significantly increased. The expression was highly positive correlation. Conclusion TGF β may promote the expression of TIMP-1, inhibit the activity of collagenase and prevent the degradation of collagen, which is an important reason for the formation of HTS.
