目的探讨黄芪注射液对大鼠脑出血灶周围凋亡神经元线粒体Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶和细胞色素C的影响。方法将32只雄性SD大鼠随机分为黄芪治疗组、出血模型组和正常对照组,其中黄芪治疗组又分为出血后6h治疗组和24h治疗组。选用胶原酶Ⅶ-肝素液注射到大鼠尾状核建立脑出血动物模型,48h后测定各组大鼠脑出血灶周围神经元线粒体Na+-K+-ATP酶和Ca2+-Mg2+-ATP酶的活性,以及神经元内细胞色素C的改变。结果与正常组比较,出血模型组细胞色素C的表达明显增加(P<0.01),ATP酶的活性均明显降低(P<0.01)。与出血模型组比较,6h治疗组及24h治疗组的细胞色素C表达均明显减少(P<0.05),ATP酶的活性均明显增加(P<0.05);6h治疗组与24h治疗组比较,细胞色素C的表达差异无统计学意义(P>0.05),ATP酶的活性6h组高于24h组(P<0.05)。结论黄芪注射液进行干预可减少脑出血灶早期神经元细胞内细胞色素C的释放及提高Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶的活性,从而减轻脑出血灶区神经细胞的凋亡,并且出血6h用药较出血24h后用药效果更好。
Objective To investigate the effect of Astragalus injection on the mitochondrial Na + -K + -ATPase, Ca2 + -Mg2 + -ATPase and cytochrome C in the apoptotic neurons around the intracerebral hemorrhage in rats. Methods Thirty-two male Sprague-Dawley rats were randomly divided into Astragalus membranaceus group, hemorrhage model group and normal control group. The astragalus membranaceus group was further divided into 6 h treatment group and 24 h treatment group. The collagenase Ⅶ -heparin solution was injected into the caudate nucleus of rats to establish the model of intracerebral hemorrhage. After 48h, the activity of mitochondrial Na + -K + -ATPase and Ca2 + -Mg2 + -ATPase in the perihepatic neurons of each group were determined, As well as neuronal cytochrome C changes. Results Compared with the normal group, the expression of cytochrome C in the hemorrhagic model group was significantly increased (P <0.01) and the ATPase activity was significantly decreased (P <0.01). Compared with the model group, the expression of cytochrome C in 6h treatment group and 24h treatment group were significantly decreased (P <0.05) and ATPase activity was significantly increased (P <0.05); Compared with 24h treatment group, There was no significant difference in the expression of pigment C (P> 0.05). The activity of ATPase in 6h group was higher than that in 24h group (P <0.05). Conclusion Astragalus injection can reduce the release of intracellular cytochrome C and increase the activity of Na + -K + -ATPase, Ca2 + -Mg2 + -ATPase in the early neuronal cells of ICH and thus reduce the apoptosis of neuronal cells in the focal cerebral hemorrhage area , And bleeding 6h medication bleeding effect better after 24h.
