目的研究抑癌基因PTEN、血管内皮生长因子(VEGF)在唾液腺腺样囊性癌(salivary adenoid cysticcarcinoma,SACC)患者体内的表达及相关性。方法选取滕州市中心人民医院病理科存档处保存的2000-2007年涎腺手术切除后的石蜡标本,其中SACC30例,多形性腺瘤(PA)26例,以正常涎腺组织10例为对照。应用免疫组化链霉菌抗生物素蛋白-过氧化酶连接(SP)法检测PTEN、VEGF在SACC、PA及正常涎腺组织中的蛋白表达。结果SACC中PTEN阳性表达率(43.33%)显著低于PA(92.30%)和正常涎腺组织(100%)(P<0.01),SACC中VEGF阳性表达率(86.67%)显著高于PA(23.09%)和正常涎腺组织(0)(P<0.01);PTEN与VEGF蛋白表达呈负相关(r=-0.40,P<0.05)。结论PTEN与VEGF在SACC的发生发展中可能起重要作用,PTEN蛋白表达的降低或缺失可能增加VEGF的表达,从而促进肿瘤的血管生成,导致肿瘤恶性进展。
Objective To study the expression and correlation of tumor suppressor gene PTEN and vascular endothelial growth factor in patients with salivary adenoid cystic carcinoma (SACC). Methods Paraffin specimens from salivary glands excised from 2000 to 2007 were collected from the Department of Pathology, People’s Hospital of Tengzhou City, including 30 cases of SACC and 26 cases of pleomorphic adenoma (PA), and 10 cases of normal salivary gland tissue as control. The protein expression of PTEN and VEGF in SACC, PA and normal salivary glands was detected by immunohistochemical streptavidin-peroxidase (SP) method. Results The positive rate of PTEN in SACC was significantly lower than that in PA (92.3%) and normal salivary gland (100%) (P <0.01). The positive expression rate of VEGF in SACC was significantly higher than that in PA (23.09% %) And normal salivary gland (0) (P <0.01). There was a negative correlation between the expression of PTEN and VEGF (r = -0.40, P <0.05). Conclusions PTEN and VEGF may play an important role in the development of SACC. The decrease or loss of PTEN protein may increase the expression of VEGF, thereby promoting tumor angiogenesis and leading to malignant progression of the tumor.
