目的:探讨人食管癌紫杉醇耐药细胞Ec9706/P-1多药耐药基因1(mdr1)、多药耐药相关蛋白基因1(mrp1)及β-微管蛋白Ⅳ基因(Hβ4)mRNA表达的意义。方法:以体外诱导的人食管癌紫杉醇耐药细胞株Ec9706/P-1为模型,采用逆转录-多聚酶链反应(RT-PCR)检测mdr1、mrp1和Hβ4在Ec9706/P-1及Ec9706中的表达变化。结果:Ec9706/P-1细胞mdr1及Hβ4 mRNA的表达高于Ec9706,差异有统计学意义(P<0.05);Ec9706/P-1及Ec9706细胞中mrp1 mRNA的表达差异无统计学意义(P>0.05)。结论:mdr1和Hβ4 mRNA高表达可能与人食管癌紫杉醇耐药细胞株Ec9706/P-1产生获得性耐药相关。
Objective: To investigate the expression of multidrug resistance gene 1 (mdr1), multidrug resistance-related protein 1 (mrp1) and β-tubulin Ⅳ (Hβ4) mRNA in human esophageal carcinoma cell line paclitaxel- significance. Methods: The human esophageal cancer cell line Ec9706 / P-1 induced by in vitro was used as a model to detect the expression of mdr1, mrp1 and Hβ4 in Ec9706 / P-1 and Ec9706 by reverse transcription polymerase chain reaction (RT-PCR) Change of expression Results: The mRNA expression of mdr1 and Hβ4 in Ec9706 / P-1 cells was higher than that in Ec9706 cells (P <0.05), while the expression of mrp1 mRNA in Ec9706 / P-1 and Ec9706 cells was not significantly different (P> 0.05). Conclusion: The overexpression of mdr1 and Hβ4 may be associated with acquired drug resistance in human esophageal cancer cell line Ec9706 / P-1.
