目的 检测Bcl- 2、Bax及c -myc在肝细胞癌中的表达 ,探讨肝细胞癌中二者表达与凋亡指数的关系。方法 对 2 2例人肝细胞癌及癌旁组织分别进行Bax及c -myc的免疫组织化学研究和TUNEL技术标记 ,计数阳性细胞数。结果 在肝细胞癌中Bax蛋白呈低表达 ,表达率为 45 45 % ,与癌旁组织相比有显著差异 (P <0 0 1) ;c -myc在肝细胞癌及癌旁组织中均呈高表达 ,表达率分别为 81 82 %和 72 73% ,但二者无差异 (P >0 0 5 ) ;肝癌组织中凋亡指数为 9 5 5 % ,低于癌旁组织15 6 8% ,二者有显著差异 (P <0 0 1)。结论 肝细胞癌的发生与细胞凋亡受到抑制有关 ,Bax起到更关键的作用 ;Bcl- 2 /Bax异二聚体能更准确地反映凋亡发生的程度 ;凋亡指数有可能成为判定细胞癌变的指标。
Objective To detect the expression of Bcl-2, Bax and c-myc in hepatocellular carcinoma and to explore the relationship between the expression of them and the apoptotic index in hepatocellular carcinoma. Methods Twenty-two cases of human hepatocellular carcinoma and paracancerous tissues were studied with Bax and c-myc immunohistochemistry and TUNEL technique. The number of positive cells was counted. Results The expression of Bax protein was low in hepatocellular carcinoma, with the expression rate of 45.5%, which was significantly different from that in the adjacent tissues (P < 0.01); c-myc was present in hepatocellular carcinoma and adjacent tissues. The high expression rate was 81 82% and 72 73%, respectively, but there was no difference between the two groups (P > 0 05). The apoptotic index in HCC tissues was 95.5%, which was lower than that of the adjacent tissue 156.8%. There was a significant difference between the two (P < 0 01). Conclusion The occurrence of hepatocellular carcinoma is related to the inhibition of apoptosis. Bax plays a more critical role; Bcl-2 / Bax heterodimer can more accurately reflect the degree of apoptosis; apoptosis index may become the judgement of cell canceration. index of.
