目的:观察通冠胶囊对动脉粥样硬化(AS)兔血管内皮功能的影响,探讨通冠胶囊抗动脉粥样硬化的作用和可能机制。方法:健康雄性新西兰白兔36只,按随机数字表法分为空白对照组、高脂对照组、阿托伐他汀组、通冠胶囊大、中、小剂量组,每组6只,空白对照组喂基本饲料,其余各组采用免疫性内皮损伤合并高脂饲料诱导建立AS兔模型,高脂对照组喂高脂饲料,阿托伐他汀组在高脂对照组的基础上加喂阿托伐他汀5mg/kg/d;通冠胶囊小、中、大剂量组在高脂对照组基础上分别加喂通冠胶囊3mg/kg/d、9mg/kg/d及15mg/kg/d,实验周期为6周,各组动物分别于2、6周末行内皮功能超声检测血管内皮舒张功能(FMD)、血管内皮非依赖性舒张功能(NMD)及血脂检测。结果:6周末,通冠胶囊大剂量组、阿托伐他汀组与高脂对照组之间FMD、NMD比较,差异均有显著性意义(P<0.05);通冠胶囊大剂量组NMD、FMD与阿托伐他汀组比较,差异无显著性意义(P>0.05)。结论:通冠胶囊能显著改善动脉粥样硬化兔血管内皮功能,其具有不依赖阿托伐他汀的独立的改善血管内皮功能的作用,而且在改善NMD方面,通冠胶囊优于阿托伐他汀。
Objective: To observe the effects of Tongguan Capsule on vascular endothelial function in atherosclerotic rabbits (AS) and explore the anti-atherosclerotic effects and possible mechanisms of Tongguan Capsule. Methods: Thirty-six healthy male New Zealand white rabbits were randomly divided into blank control group, high fat control group, atorvastatin group, Tongguan capsule large, medium and small dose group, 6 rats in each group, blank control The other groups were fed with high-fat diet and high-fat diet. Endothelium-dependent ultrasound was used to detect the endothelium-dependent vasodilation (FMD), endothelium-independent vasodilatation (NMD) and serum lipids in each group at 2 and 6 weeks. Results: There was significant difference in FMD and NMD between Tongguan capsule high-dose group, atorvastatin group and high-fat control group at 6weeks (P <0.05); Tongguan capsule high-dose group NMD, FMD Compared with atorvastatin group, there was no significant difference (P> 0.05). Conclusion: Tongguan Capsule can significantly improve the endothelial function of atherosclerotic rabbits, which has the independent role of ameliorating vascular endothelial function independent of atorvastatin. Tongguan Capsule is superior to atorvastatin in improving NMD .
