目的建立同时测定苦豆子药材中金雀花碱、苦参碱、氧化苦参碱、槐果碱、氧化槐果碱和槐定碱的RP-HPLC方法,测定不同产地苦豆子药材中不同部位6种生物碱的含量。方法色谱柱为Kromasil C18(4.6 mm×250 mm,5μm);流动相为甲醇(A)-0.2%磷酸水(B);洗脱梯度为:0 min,3%A;20 min,5%A;40 min,6%A;60 min,7%A;紫外检测波长为215 nm,流速为1 m L·min-1,柱温30℃。结果 6种生物碱的线性关系良好(r≥0.999 3),平均回收率为97%~104%。各产地苦豆子样品药材中果实部位的6种生物碱含量都明显高于根、茎、叶、壳部位中各碱的含量;宁夏、甘肃的药材果实部位中主要生物碱含量稍高于其他地区。结论该方法简便、准确、快速,适用于苦豆子生物碱的含量测定。
OBJECTIVE To establish a RP-HPLC method for the simultaneous determination of serovar, matrine, oxymatrine, sophocarpine, oxycinnamine and sophoridine in Sophora alopecuroides, and to determine the content of different parts of Sophora alopecuroides in different areas Alkaloid content. Methods The chromatographic column was Kromasil C18 (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol (A) -0.2% phosphoric acid water (B). The elution gradient was 0 min, 3% A, 20 min, 5% ; 40 min, 6% A; 60 min, 7% A; UV detection wavelength of 215 nm, flow rate of 1 m L · min-1, column temperature 30 ℃. Results The linear relationship between the six alkaloids was good (r≥0.9993) and the average recovery was 97% -104%. The contents of 6 alkaloids in the fruit parts of Sophora alopecuroides samples from all regions were significantly higher than those from roots, stems, leaves and shell parts. The contents of major alkaloids in the fruit parts of Ningxia and Gansu were slightly higher than those in other regions . Conclusion The method is simple, accurate and rapid and suitable for the determination of Alkaloids of Sophora alopecuroides.
