目的对本课题组合成的氨基酸螯合钙进行药效学评价,为进一步研发奠定基础。方法不同剂量的氨基酸螯合钙对去势大鼠进行30 d的治疗并使用阳性药复方氨基酸螯合胶囊和空白制剂作为对照。通过大鼠骨代谢生化指标和骨力学指标评价氨基酸螯合钙药效学。结果去势组大鼠子宫系数显著下降(P<0.01)。氨基酸螯合钙能够显著降低去势大鼠血中的碱性磷酸酶水平,提高血钙水平(P<0.05)。与模型对照组比较,氨基酸螯合钙组在增加股骨体积、股骨灰重、干重、股骨极限压应力和极限剪力方面存在显著性的差异(P<0.05)。结论本课题组合成的氨基酸螫合钙在改善去势大鼠骨质疏松方面具有一定的作用,为该制剂进一步研究和优化提供了实验依据。
OBJECTIVE To evaluate the pharmacodynamics of amino acid chelated calcium synthesized by this research group, and lay the foundation for further research and development. Methods Different doses of calcium amino acid chelate were administered to ovariectomized rats for 30 days and the positive drug compound amino acid chelating capsules and blank preparations were used as control. Evaluation of Amino Acid Chelating Calcium Pharmacodynamics by Biochemical and Biomechanical Indexes of Bone Metabolism in Rats. Results Ovariectomy rats significantly decreased (P <0.01). Amino acid chelated calcium can significantly reduce the alkaline phosphatase level and increase the level of serum calcium in ovariectomized rats (P <0.05). Compared with the model control group, the amino acid chelated calcium group had significant difference in increasing femoral volume, femoral gray weight, dry weight, ultimate compressive stress and ultimate shear force of femur (P <0.05). Conclusion This research group synthesized calcium amino acid in the improvement of ovariectomized rats has a role in osteoporosis, and provide experimental evidence for the further study and optimization of the preparation.
