目的:对阴茎海绵体平滑肌细胞纤维化模型进行不同浓度红景天苷干预以找出最适的抗纤维化浓度;方法:低氧(1%O2浓度)48h诱导阴茎海绵体平滑肌细胞纤维化;分别加入前列腺素E1(PGE1)0.4μg/L、红景天苷1、3、5 mg/L,未加药物组作为对照,RT-PCR分别测定各组TGF-β1、Ⅰ型胶原、Ⅲ型胶原的相对表达量。结果:RT-PCR结果提示红景天苷和前列腺素E1都能起到不同程度的抗纤维化作用,其中红景天苷3 mg/L与前列腺素E1的抗纤维化作用无显著差异(P>0.05),增加红景天苷的浓度不能增强其作用效果。结论:红景天苷3mg/L在体外抗阴茎海绵体平滑肌细胞纤维化的作用效果与经典药物前列腺素E1相当,可为进一步临床研究阴茎海绵体纤维化提供理论支持。
OBJECTIVE: To investigate the effect of salidroside at different concentrations on the fibrosis model of corpus cavernosum smooth muscle cells in order to find out the optimal anti-fibrosis concentration. Methods: The hypoxia (1% O2) induced the fibrosis of smooth muscle cells of the corpus cavernosum for 48 hours. (PGE1) 0.4μg / L, salidroside 1,3,5 mg / L, respectively, without drug group as control, RT-PCR were determined TGF-β1, type Ⅰ collagen, type Ⅲ The relative expression of collagen. Results: RT-PCR results suggest that salidroside and prostaglandin E1 can play different degrees of anti-fibrosis, including salidroside 3 mg / L and prostaglandin E1 anti-fibrosis no significant difference (P > 0.05), increase the concentration of salidroside can not enhance its effect. Conclusion: The effect of salidroside 3mg / L on anti-fibrosis of penis corpus cavernosum in vitro is comparable with that of classical drug prostaglandin E1, which may provide theoretical support for further clinical study of penis fibrosis.
