在以快速而准确统计染色体数目为目的的甜菜细胞压片制片中,秋水仙碱、8—羟基喹啉以及对二氯苯是实验室最常用的预处理化学药剂。结合冷冻,Rosen G.用秋水仙碱(1949),БормотовВ.Е.等用8—羟基喹啉(1966),李盛贤等用对二氯苯(1982)均得到积累中期相染色体的良好制片结果。然而作者们没有比较冷冻以及冷冻与这些化学药剂分别结合的不同处理之间,在制片效果上的异同,也没有探讨不同预处理积累中期相的作用机理。本文观察并讨论了不同预处理对甜菜染色体引起的形态变化,并比较其制片效果的异同,初步分析了对细胞分裂周期引起的变比。
Colchicine, 8-hydroxyquinoline, and p-dichlorobenzene are the most commonly used pretreatment chemistries in beet cell pellets for the purpose of rapid and accurate counting of chromosome numbers. Combined with freezing, Rosen G. using colchicine (1949), Бормотов В.Е. With 8-hydroxyquinoline (1966), Li Shengxian and other use of p-dichlorobenzene (1982) were obtained good metaphase chromosome production results . However, the authors did not compare the similarities and differences between the different treatments of freezing and freezing and the combination of these chemicals, respectively, and did not discuss the mechanism of action of different pretreatments to accumulate metaphase. In this paper, we observed and discussed the morphological changes caused by different pretreatment on chromosomes of beet and compared the similarities and differences in the production of sugar beet. The transformation ratio caused by the cell cycle was analyzed preliminarily.
