文章对海洋真菌碱提多糖YCC进行了初步结构鉴定,并探讨了其对人肾小球系膜细胞(MCs)增殖的作用。采用了气相色谱法、红外光谱法、高碘酸氧化法、Smith降解等方法测定了多糖的结构。用四甲基偶氮唑盐(MTT)法检测,观察了YCC对高糖培养的MCs增殖的影响。结果表明,YCC的平均相对分子质量为66k,单糖组成为:甘露糖、葡萄糖、半乳糖,其摩尔比为1.8∶81.33∶16.87;比旋度测定及IR结果表明,YCC为α构型;高碘酸氧化及Smith降解的结果显示,YCC的连接方式主要为1→6连接、1→4连接和1→3连接。YCC浓度在1mg/ml时对高糖所致的MCs增殖有显著的抑制作用。
In this paper, the preliminary structural elucidation of marine fungal alkaline polysaccharide YCC was carried out and its effect on the proliferation of human mesangial cells (MCs) was explored. The structures of polysaccharides were determined by gas chromatography, infrared spectroscopy, periodate oxidation and Smith degradation. The effect of YCC on proliferation of MCs cultured in high glucose was observed by MTT assay. The results showed that the average relative molecular mass of YCC was 66k, the composition of monosaccharides was mannose, glucose and galactose, the molar ratio was 1.8: 81.33: 16.87. The specific rotation and IR results showed that YCC was α - The results of periodate oxidation and Smith degradation showed that the connection modes of YCC mainly consisted of 1 → 6 connections, 1 → 4 connections and 1 → 3 connections. YCC concentration of 1mg / ml of high glucose-induced MCs proliferation was significantly inhibited.
