环孢素(Cyclosporine A,CsA)是一种重要的免疫抑制剂药物。它用于器官移植,可延长移植后器官的成活率、预防移植器官的排斥反应,但CsA的血药浓度与疗效和毒副反应密切相关。监测血药浓度对提高安企有效的用药至关重要。 目前CsA的测定方法有RIA、萤光偏振免疫(TDX)、放免单克隆抗体等法。但这些方法易受代谢物影响,产生交叉反应,且分析精度也不如HPLC法好。用HPLC法测定CsA血药浓度国外已有报道。但几乎都用C_8或C_(18)柱分析,以乙腈或甲醇与水二元溶剂为流动相,使用柱温(70~79℃)。国内对CsA的分析方法报道甚少,并选用C_(18)或C_8柱。本文选用极性较大的CN
Cyclosporine A (CsA) is an important immunosuppressant drug. It is used in organ transplantation to prolong the survival rate of organs after transplantation and prevent the rejection of transplanted organs. However, the plasma concentration of CsA is closely related to the curative effect and side effects. Monitoring blood concentration is essential to improve the effective use of drugs. The current determination of CsA RIA, fluorescence polarization immunity (TDX), RIA monoclonal antibody and other methods. However, these methods are susceptible to metabolites, produce cross-reactions, and are less analytical than HPLC. HPLC method for the determination of CsA blood concentration abroad has been reported. But almost all with C_8 or C_ (18) column analysis, acetonitrile or methanol and water binary solvent as the mobile phase, the use of column temperature (70 ~ 79 ℃). There are few reports about CsA in China and C_ (18) or C_8 columns have been selected. In this paper, choose a more polar CN
