WHO建议,对生产狂犬病疫苗细胞系的纯度以及病毒株的抗原有无改变要定期进行评估。此文作者用SDS-PAGE和考马斯/银染法分析了17种市售人狂犬病疫苗的病毒及非病毒蛋白。富含的60~70 kD信号肽的质谱分析表明,在多数疫苗中人源性血清白蛋白(HSA)是主要成分,在所有检测的PVRV(Vero细胞狂犬病纯化疫苗)、HDCV(人二倍体细胞狂犬病疫苗)和PHK(原代地鼠肾)疫苗中,1剂平均含有22 mg的HSA。与之相反,在PCECV(鸡胚细胞培养的纯化疫苗)和PDEV(鸭胚狂犬病疫苗)中HSA浓度分别为前者的1/1000和测不出。
WHO recommends that periodic evaluation be performed on the purity of the rabies vaccine cell line and on the presence or absence of antigen in the virus strain. The authors analyzed the viral and non-viral proteins of 17 commercially available human rabies vaccines by SDS-PAGE and Coomassie / silver staining. Mass spectrometric analysis of the enriched 60-70 kD signal peptide showed that in most vaccines human serum albumin (HSA) is the predominant component of all the tested PVRV (Vero Cell Rabies Purified Vaccine), HDCV (human diploid Cell rabies vaccine) and PHK (primary murine kidney) vaccine, one dose contains on average 22 mg of HSA. In contrast, the HSA concentrations in PCECV (chick embryo cultured purified vaccine) and PDEV (duck embryo rabies vaccine) were 1/1000 and undetectable, respectively.
