为探索空间条件对植物种子的诱变作用,对粳稻“农垦58”纯系单株种子经“8885”卫星搭载处理后在地面种植,筛选出已稳定遗传的大粒型突变系,对其基因组的突变位点进行了分子生物学分析,运用140个随机引物对“农垦58”原种及其大粒型突变系进行多态性研究,在扩增的2000多条DNA片段中,仅有5个片段显示了多态性,即所检测的2000个染色体位点上仅5个位点有差异,占0.25%,其中4个片段为重复序列,一个片段为2个拷贝序列。应用水稻窄叶青8号和京系17杂交F1代花药培养DH群体为作图群体,将该片段定位于第11号染色体上,位于分子标记CDO520和PTA818中间。
In order to explore the mutagenic effects of space conditions on plant seeds, the single-line seed of japonica “Nongken 58” was planted on the ground after being sown by “8885” satellites and the stable mutant large grain-type mutant line was screened. The mutation sites were analyzed by molecular biology. 140 random primers were used to study the polymorphism of the “Nongken 58” and its large grain mutant lines. Only 5 of the more than 2000 DNA fragments amplified The results showed that only 5 sites on the 2,000 chromosomal loci detected had a difference of 0.25%, of which 4 were duplicates and one was 2 copies. The DH population was obtained from the F1 anthers of rice hybrids ZYYP 8 and Jing 17 by mapping populations. The fragment was located on chromosome 11 and located between the molecular markers CDO520 and PTA818.
