[Objective] To study the polysaccharides compositions in ripe peel of Citrus reticulate ’Chachi’.[Method] Employing HPLC method,PMP pre-column derivatization was performed to measure the monosaccharide composition of pectic polysaccharides of Citrus reticulate ’Chachi’,under the chromatographic conditions as follows:chromatographic columns of the Phenomenex Luna C18(250 mm×4.60 mm,5 μm),with the mobile phases of 0.05 mol/L mixture solution of phosphates-acetonitrile(80:20,V/V,pH=7.1),flow rate of 1.0ml/min;detection wavelength of 245 nm,column temperature of 30 ℃ and injection volume of 20 μl.[Result] The pectic polysaccharides of Citrus reticulate ’Chachi’ contained mannose,ribose,rhamnose,galacturonic acid,glucose,galactose,xylose,arabinose and so on.The mole ratios of each monosacchavide was correspondingly 0.08:0.002:0.05:4.80:1:1.52:0.11:3.82.[Conclusion] This method has the advantages of simple operation,shortened period,repeatability and yield,and could be used for the measurement of polysaccharides of Citrus reticulate ’Chachi’.
