目的探讨孕期低强度跑台运动对宫内高糖干预的子代雄鼠血管内皮功能的影响。方法采用Sprague-Dawley新生鼠链脲佐菌素腹腔注射造成一过性高血糖,建立妊娠期糖尿病(gestational diabetes mellitus,GDM)模型,GE组行孕期低强度跑台运动(坡度0°,速度10m/min,持续10min),检测孕晚期母鼠氧化应激状态及血糖水平;彩色超声多普勒检测8周龄子代雄鼠弹丸式注射乙酰胆碱和舌下含服硝酸甘油前后腹主动脉内径变化。结果孕21 d,GDM组血糖显著升高,且高于GE组(P<0.05);8周龄子鼠,GDM组与血糖水平均增高(P<0.05)且两组间未见明显差异(P>0.05)。GDM组超氧化物歧化酶活性降低,且丙二醛浓度升高;GE组完全逆转两者改变(P<0.05)。GDM组子鼠腹主动脉内膜轻度增厚,EDD显著降低;GE组较GDM组明显升高(P均<0.05)。各组腹主动脉EID未见明显差异(P>0.05)。结论孕期低强度跑台训练完全逆转GDM相关的氧化抗氧化失衡,部分改善子代成年期血管内皮功能。
Objective To investigate the effects of low intensity treadmill exercise during pregnancy on the vascular endothelial function of offspring in offspring of intrauterine hyperglycemia. Methods The model of gestational diabetes mellitus (GDM) was established by intraperitoneal injection of streptozotocin (STZ) in Sprague-Dawley newborn rats. Gestational diabetes mellitus (GE) / min for 10 min). The changes of oxidative stress and blood glucose level in the third trimester of pregnant women were detected by color Doppler sonography. The changes of the internal diameter of the abdominal aorta of the 8-week-old progeny male mice after bolus injection of acetylcholine and sublingual nitroglycerin . Results The level of blood glucose in GDM group was significantly higher than that in GE group (P <0.05) at 21 d of gestation. There was no significant difference between the two groups (P <0.05) P> 0.05). The activity of superoxide dismutase (SOD) in GDM group decreased and the concentration of malondialdehyde (MDA) increased. In GE group, the changes were both reversed (P <0.05). The intimal thickening of abdominal aorta in GDM group was mild and the EDD was significantly lower than that in GDM group (all P <0.05). There was no significant difference in EID of abdominal aorta in each group (P> 0.05). Conclusion Low-intensity treadmill training during pregnancy completely reversed the imbalance of oxidative and anti-oxidative activities associated with GDM, and partially improved the vascular endothelial function in offspring in adulthood.
