四种心肌保护方法对心肌细胞质膜蛋白的影响

目的:研究4种心肌保护方法对心肌细胞质膜蛋白结构和功能的影响,评价其心肌保护效果。方法:复制动物模型,检测心肌细胞质膜 ATP酶活性、质膜蛋白形态、二级结构及电泳谱型变化。结果: (1)缺血期间,各组 Na~+,K~+泵活性均显著下降;体外循环 180 min, Ⅵ组恢复较好。 Ca~(2+),Mg~(2+)泵与 Na~+,K~+泵活性变化规律相似。(2)复灌后1. 8万以下质膜蛋白条带出现不同程度缺失。(3)各组间质膜蛋白颗粒等效直径无明显差异。缺血期间,Ⅵ组单位面积质膜蛋白颗粒数无明显减少。(4)各组心肌细胞质膜总蛋白二级结构呈无规律变化。结论:单纯 CPB对心肌细胞质膜蛋白无影响;激活的蛋白酶在缺血再灌注损伤中可能具有重要作用;4种心肌保护方法均不能完全遏制再灌注损伤的发生;温血停搏液心肌保护效果较好。 Objective: To study the effects of four kinds of myocardial protection methods on the plasma membrane protein structure and function of cardiomyocytes, and to evaluate the myocardial protection effect. Methods: The animal models were duplicated and the changes of plasma membrane ATPase activity, plasma membrane protein morphology, secondary structure and electrophoretic profile were examined. Results: (1) During the ischemic period, the Na + and K + pump activities in each group were significantly decreased. The extracorporeal circulation was 180 min, and the recovery in group Ⅵ was better. The activities of Ca ~ (2 +), Mg ~ (2+) pump and Na ~ +, K ~ + pump were similar. (2) After reperfusion 1. The following 80,000 plasma membrane protein bands appear to varying degrees of deletion. (3) There was no significant difference in equivalent diameter of plasma membrane protein between groups. During ischemia, there was no significant decrease in the number of plasma membrane protein per unit area in group VI. (4) The secondary structure of plasma membrane total protein in each group showed irregular changes. CONCLUSION: CPB alone has no effect on the plasma membrane protein of myocardium. The activated protease may play an important role in ischemia-reperfusion injury. All four kinds of myocardial protection methods can not completely restrain the occurrence of reperfusion injury. The cardioprotective effects of warm cardioplegia better.
