苯丁酸氮芥是一种烷化剂,有广谱抗肿瘤作用。美国西南肿瘤组曾在小规模研究中,单用本药使12例组织细胞增生症 X 患者中9例完全缓解(CR;占75%),2例部份缓解(PR),仅1例无效。而以往用单一药物,如长春新碱(VCR)、环磷酰胺(CTX),或 Velban,则有效率(CR+PR)仅为50%左右。由于患组织细胞增生症 X 病儿对联合化疗反应欠佳,故进一步设计本研究以确定单用苯丁酸氮芥的效果。作者选择20岁以下、经活检证实之组织细胞增生症 X 患者82例,其中不包括儿童嗜酸性肉芽肿。将患
Chlorambucil is an alkylating agent with a broad spectrum of anti-tumor effects. In a small-scale study, the Southwest Oncology Group of the United States used 9 cases of complete remission (CR; 75%), 2 partial response (PR), and only 1 case of complete remission in 12 patients with histiocytosis X . Previously, single agents such as vincristine (VCR), cyclophosphamide (CTX), or Velban were found to have an effective rate (CR + PR) of only about 50%. Because of the poor response of patients with histiocytosis X to combined chemotherapy, the study was further designed to determine the efficacy of chlorambucil alone.
