为观察硝苯地平和依那普利对兔髂动脉血管成形术后胶原增生的作用 ,将 32只雄性新西兰大白兔随机分为对照组、硝苯地平组和依那普利组 ,行右髂总动脉血管成形术。硝苯地平组和依那普利组于术前一天分别从耳静脉注射硝苯地平 10 0ng g和依那普利 0 .4μg g,每 12h一次 ,至术后两周。分别测定术前、术后 30min及术后两周血浆血管紧张素Ⅱ含量 ,行Masson’s三色染色并进行图像分析。结果发现 ,对照组术后 30min血管紧张素Ⅱ含量由 2 .70± 0 .31g L升高为 2 .83± 0 .18g L (P <0 .0 5 ) ,两周后恢复至术前水平 ( 2 .5 0± 0 .2 6g L ,P >0 .0 5 ) ;硝苯地平组血管紧张素Ⅱ含量与对照组无明显差异。依那普利组术后 30min血管紧张素Ⅱ含量为 2 .5 0± 0 .0 9g L ,较术前 ( 2 .5 7± 0 .10g L)无升高 (P >0 .0 5 )。图像分析胶原含量表明 ,血管损伤后两周血管壁胶原含量增高 ,面密度、平均光密度及积分光密度明显增高 (P <0 .0 1) ;依那普利组面密度、平均光密度及积分光密度显著低于对照组 (P <0 .0 1)。结果提示 ,血管成形术后血管壁发生明显的胶原增生 ,依那普利抑制血管壁胶原增生 ,可能对预防经皮腔内冠状动脉成形术后再狭窄有效
To observe the effect of nifedipine and enalapril on collagen hyperplasia in rabbit after iliac artery angioplasty, 32 male New Zealand white rabbits were randomly divided into control group, nifedipine group and enalapril group. The right iliac Total arterial angioplasty. Nifedipine group and enalapril group were injected with nifedipine 10 ng g and enalapril 0.4 μg g from the ear vein on the day before surgery, once every 12 hours until two weeks after operation. The plasma levels of angiotensin Ⅱ were measured before operation, 30min after operation and two weeks after operation respectively. Masson’s trichrome staining and image analysis were performed. The results showed that the level of angiotensin Ⅱ in the control group increased from 2.70 ± 0.31g L to 2.83 ± 0.18g L (P <0.05) at 30min after operation, and returned to preoperative level after two weeks (2.5 ± 0.2 6g L, P> 0.05). The content of angiotensin Ⅱ in nifedipine group was not significantly different from that in control group. The concentration of angiotensin Ⅱ in enalapril group was 2.50 ± 0.99g at 30min after operation, which was not significantly higher than that of preoperative (2.57 ± 0.10g L) (P> 0.05) . Image analysis of collagen content showed that two weeks after blood vessel injury, vascular wall collagen content increased, surface density, average optical density and integral optical density increased significantly (P <0.01); enalapril group surface density, average optical density and The integral optical density was significantly lower than that of the control group (P <0.01). The results suggest that significant collagen hyperplasia occurs on the vessel wall after angioplasty and that enalapril inhibits the proliferation of vascular wall collagen may be effective in preventing restenosis after percutaneous transluminal coronary angioplasty
