目的:优选黄连总生物碱明胶微球的制备工艺。方法:采用乳化交联法制备黄连总生物碱明胶微球,以包封率和载药量的综合评分为指标,通过L9(34)正交试验考察明胶质量分数、投药量、交联剂用量与水油比对制备工艺的影响。结果:最佳制备工艺为明胶质量分数12%,投药量1.7%,交联剂用量15%,水相油相比1∶12。载药微球呈圆球形,表面光滑,包封率63.73%,载药量6.50%。结论:优化的制备工艺稳定可行,制备的微球质量较好。
Objective: To optimize the preparation of Coptis alkaloids gelatin microspheres. METHODS: The total alkaloids of Coptidis Rhizoma coptis gelatin were prepared by emulsification and cross-linking method. The comprehensive evaluation of encapsulation efficiency and drug loading was taken as the index. The gelatin mass fraction, dosage, crosslinker dosage And water to oil ratio on the preparation process. Results: The optimal preparation process was gelatin 12%, dosage 1.7%, crosslinking agent 15% and water phase oil 1:12. Drug-loaded microspheres were spherical, smooth surface, encapsulation efficiency 63.73%, drug loading 6.50%. Conclusion: The optimized preparation process is stable and feasible, and the prepared microspheres have better quality.
