[目的]研究冷冻治疗对荷瘤Wistar大鼠生存期的影响和机制。[方法]建立大鼠C6脑胶质瘤模型,冷冻治疗后免疫组化检测p27kip1和CyclinD1蛋白的表达,观察动物生存期。[结果]与未实施冷冻治疗对照组比较,冷冻治疗可延长动物生存期,上调p27kip1蛋白的表达,下调CyclinD1蛋白的表达。p27kip1蛋白阳性表达率与荷瘤鼠生存期呈正相关,CyclinD1蛋白的阳性表达率与荷瘤鼠生存期呈负相关。p27kip1蛋白阳性表达率和CyclinD1蛋白的阳性表达率呈负相关。[结论]冷冻治疗可延长动物生存期,其机制同上调p27kip1蛋白和下调CyclinD1蛋白的表达有关。
[Objective] To investigate the effect and mechanism of cryotherapy on the survival of Wistar rats with tumor. [Method] The model of rat C6 glioma was established. After frozen treatment, the expressions of p27kip1 and CyclinD1 protein were detected by immunohistochemistry and the survival of animals was observed. [Results] Compared with the control group without cryotherapy, cryotherapy could prolong the survival of animals, up-regulate the expression of p27kip1 protein and down-regulate the expression of CyclinD1 protein. The positive rate of p27kip1 expression was positively correlated with the survival of tumor-bearing mice. The positive rate of CyclinD1 protein expression was negatively correlated with the survival of tumor-bearing mice. The positive rate of p27kip1 protein was negatively correlated with the positive rate of CyclinD1 protein. [Conclusion] Cryotherapy can prolong the survival of animals. Its mechanism is related to up-regulation of p27kip1 protein and down-regulation of the expression of CyclinD1 protein.
