目的 探讨抑癌基因、细胞周期调控与胶质瘤发生、发展的关系 .方法 采用免疫组织化学技术对 6 3例脑胶质瘤组织及 10例正常脑组织标本中 p2 1WAF1 /CIP1与 p5 3基因的表达状况进行检测 ,并进行相关性分析 .结果 110例正常脑组织均为 p2 1WAF 1 /CIP1阳性表达 ,6 3例脑胶质瘤组织中p2 1WAF1 /CIP1的表达阳性率与肿瘤分化程度呈正相关 ;2 10例正常脑组织中仅 1例为突变型 p5 3弱阳性表达 .脑胶质瘤组织中突变型 p5 3的阳性率与分化程度呈负相关 ;3大部分突变型 p5 3表达阳性标本中 p2 1WAF 1 /CIP1为阴性表达 ,而大部分突变型 p5 3表达阴性的标本中 p2 1WAF 1 /CIP1 为阳性表达 .结论 p2 1WAF1 /CIP1的突变或缺失可能与胶质瘤的发生、发展有关 ,p2 1WAF1 /CIP1表达的减弱或消失可能是由 p5 3突变引起 ,野生型p5 3基因的肿瘤抑制作用可能是通过 p2 1WAF 1 /CIP1 而实现
Objective To investigate the relationship between tumor suppressor genes and cell cycle regulation and the occurrence and development of gliomas.Methods Immunohistochemistry was used to detect the expression of p21WAF1 / CIP1 and p5 3 genes in 6 3 glioma tissues and 10 normal brain tissues And the correlation analysis was performed.Results The positive expression rate of p2 1WAF 1 / CIP1 in 110 normal brain tissues was positive.The positive rate of p21WAF1 / CIP1 expression in 63 glioma tissues was positively correlated with the degree of tumor differentiation , And only 1 of 210 cases of normal brain tissue showed a weak positive expression of mutant p5 3. The positive rate of mutant p5 3 in glioma tissue was negatively correlated with the degree of differentiation.3 most of the mutant p5 3 positive The expression of p2WAF1 / CIP1 in the specimens was negative, while the expression of p2WAF1 / CIP1 in most specimens with negative expression of p53 was positive.Conclusion The mutation or deletion of p2WAF1 / CIP1 may be associated with the occurrence and development of gliomas , The decrease or disappearance of p2 1WAF1 / CIP1 expression may be caused by the mutation of p5 3, and the tumor inhibition of wild type p5 3 gene may be achieved by p2 1WAF 1 / CIP1
