气相色谱-质谱是一种能够对药物及其代谢物进行分离和鉴定的方法,然而该方法需要非常困难、繁琐的衍生化和清洗过程。超高效液相色谱-质谱 (UHPLC-MS) 联用技术突破了以上局限并实现了更加快速的分析效果。本文讨论了如何通过对UHPLC-MS方法中固定相、流动相以及检测器方面的优化,在12 min的分离时间内实现了14种违禁药物及其代谢物在pg级水平上的定量。这些违禁药物及其代谢物使用Hypersil GOLD PFP (perfluorophenyl)色谱柱(1.9 μm, 100 mm×2.1 mm)进行分离,并采用单四极杆质谱快速扫描检测。
Gas chromatography-mass spectrometry is a method that separates and identifies drugs and their metabolites. However, this method requires very difficult and cumbersome derivatization and cleaning processes. UHPLC-MS technology breaks through these limitations and enables faster analysis. This article discusses how to quantify pg levels of 14 illicit drugs and their metabolites over a 12-min separation time by optimizing the stationary phase, mobile phase, and detector in the UHPLC-MS method. The banned drugs and their metabolites were separated on a Hypersil GOLD PFP (perfluorophenyl) column (1.9 μm, 100 mm × 2.1 mm) and detected using a single quadrupole mass spectrometry rapid scan.
