目的探讨肿瘤热休克蛋白70-多肽复合物(heat shock protein 70-peptide complexes,HSP70-PCs)负载的树突状细胞(dendritic cells,DCs)对肺癌细胞(SPC-A-1)的杀伤效果,并与未加抗原的DCs进行比较。方法用加热、ADP亲和层析法提取肺癌细胞HSP70-PCs作为抗原,使之与DCs共同培养,检测其对肺癌细胞的杀伤作用。结果与未加抗原的DCs比较,加入HSP70-PCs的DCs对肺腺癌细胞SPC-A-1的杀伤效果明显升高。结论负载HSP70-PCs的DCs对肺腺癌细胞SPC-A-1的杀伤效果明显高于未加抗原的DCs。
Objective To investigate the killing effect of dendritic cells (DCs) loaded with tumor heat shock protein 70-peptide complexes (HSP70-PCs) on lung cancer cells (SPC-A-1) And compared with non-antigen DCs. Methods The HSP70-PCs of lung cancer cells were extracted by heat and ADP affinity chromatography and cultured with DCs to detect the killing effect on lung cancer cells. Results Compared with DCs without antigen, the killing effect of DCs containing HSP70-PCs on SPC-A-1 cells was significantly increased. Conclusion The killing effect of DCs loaded with HSP70-PCs on SPC-A-1 cells was significantly higher than that of DCs without antigens.
