本文讨论了肝组织和血清中维生素E含量测定过程中皂化条件和维生素A对测定结果的影响。 (1)皂化时KOH用量与测定时生育酚乙酸盐含量的关系:观察到用10NKOH0.5ml皂化时,测定管中纯生育酚乙酸盐的量在0.5-100μg之间与其荧光强度呈线性关系;大于150μg时,荧光强度不随其量增加。当测定管中仅有5μg纯生育酚乙酸盐时,10NKOH的用量加到3ml不影响结果。 (2)样品量对皂化效果的影响,用10NKOH0.5ml皂化时:肝组织的用量不得超过0.1g,否则结果会偏低。测定血清时,由于其中的维生素E主要是游离型的,无须皂化。有时皂化反而造成结果降低。 (3)维生素A对维生素E含量测定的干扰:维生素A有“熄灭荧光”的作用,本研究中观察到2μg维生素E中若有0.5IU以上的维生素A时,荧光强度显著下降。在苹取维生素E已烷液中加60%硫酸0.6ml可消除其影响。但硫酸加到1.5ml时也会使结果偏低,故在用硫酸消除维生素A干扰时,也应注意硫酸用量。
This article discusses the effects of saponification and vitamin A on the determination of vitamin E in liver tissue and serum. (2) the impact of sample size on the saponification effect, saponification with 10NKOH0.5ml: the amount of liver tissue should not exceed 0.1g, otherwise the result will be low. Determination of serum, due to which the vitamin E is mainly free-type, without saponification. Sometimes saponification results in reduced results. (3) The interference of vitamin A on the determination of vitamin E: Vitamin A has the function of “extinguishing fluorescence”. In this study, it was observed that the fluorescence intensity of 2 μg of vitamin E was significantly decreased if it was above 0.5 IU. In the extraction of vitamin E hexane solution plus 60% sulfuric acid 0.6ml eliminate its effect. However, when added to 1.5ml of sulfuric acid will result in low, so the use of sulfuric acid to eliminate the interference of vitamin A, it should also pay attention to the amount of sulfuric acid.
