目的 研究nm2 3 H1基因表达与胰腺癌淋巴结转移及细胞增殖状态的关系。方法 用免疫组织化学S P法 ,检测 2 9例胰腺癌标本中nm2 3 H1基因产物和增殖细胞核抗原 (PCNA)的表达状况 ,并应用图像分析技术检测胰腺癌细胞核DNA含量及DNA倍体构成。结果 在nm2 3 H1高表达的 11例胰腺癌中 ,伴有淋巴结转移 6例 ,而nm2 3 H1低表达者 18例中仅 2例伴淋巴结转移 ,两者比较 ,P <0 0 5 ;nm2 3 H1表达与肿瘤部位、大小、组织学类型及患者性别等因素无相关 (P >0 0 5 ) ;nm2 3 H1高、低表达的胰腺癌中PCNA表达指数分别为 (2 6± 14) %和 (10± 5 ) % ,两者相比差异有非常显著意义 (P <0 0 1) ;细胞核DNA指数分别为 (2 2 5± 0 5 6 )和 (1 6 7± 0 2 3) ,两者相比差异有非常显著意义 (P <0 0 1) ;在DNA倍体分布形式上 ,nm2 3 H1高表达者二倍体细胞检出率低 (P <0 0 5 ) ,而非整倍体细胞检出率高 (P <0 0 1)。结论 nm2 3 H1基因产物过度表达与胰腺癌淋巴结转移及高细胞增殖活性有关。
Objective To investigate the relationship between nm23 H1 gene expression and lymph node metastasis and cell proliferation in pancreatic cancer. Methods Immunohistochemical SP method was used to detect the expression of nm23 H1 gene and proliferating cell nuclear antigen (PCNA) in 29 pancreatic cancer specimens. The DNA content and DNA ploidy of pancreatic cancer cells were detected by image analysis. Results In 11 cases with high expression of nm23 H1N1, 6 cases were associated with lymph node metastasis, while only 2 cases with lymph node metastasis were found in 18 cases with low expression of nm23 H1, P <0 05; nm 2 3 The expression of H1 was not correlated with tumor location, size, histological type and patient’s sex (P> 0.05). The expression of PCNA in high and low expression of nm23 H1 was (26 ± 14)% and (10 ± 5)%, respectively. There was significant difference between the two groups (P <0.01). The nuclear DNA indexes were (2 2 5 ± 0 5 6) and (1 6 7 ± 0 2 3) (P <0.01). In terms of DNA ploidy distribution, the detection rate of diploid cells with high expression of nm23 H1 was low (P <0.05), but not with aneuploidy The detection rate of somatic cells was high (P <0.01). Conclusion The overexpression of nm23 H1 gene is associated with lymph node metastasis and high cell proliferation in pancreatic cancer.
