目的:测定蜈蚣三七总皂苷中有效成分W3在大鼠体内的药代动力学参数。方法:采用反相高效液相色谱(RP-HPLC)法,大鼠ig给药后,以蜈蚣三七提取物中主要有效成分W3为指标进行测定。结果:W3血药浓度在1.56~50mg/L范围内线性关系良好,回归方程为Y=5.85X+11.69,r=0.9987(n=3);W3三种浓度的平均回收率及相对标准偏差分别为86.44%(4.22%),92.48%(5.84%),81.97%(9.32%);W3在大鼠体内的药代动力学过程符合一室吸收模型,主要药动学参数为t1/2=(2.99±0.35)h,T(peak)=(2.14±0.27)h。结论:建立了蜈蚣三七有效成分W3血药浓度的HPLC测定方法,阐明了蜈蚣三七提取物的药动学参数,方法的专属性强,结果可靠。
Objective: To determine the pharmacokinetic parameters of active ingredient W3 in total saponins of Panax notoginseng in rats. METHODS: After ig administration in rats, RP-HPLC method was used to determine the main active ingredient W3 in the extract of Radix Notoginseng. RESULTS: The W3 plasma concentration was linear in the range of 1.56-50 mg/L. The regression equation was Y=5.85X+11.69, r=0.9987 (n=3). The average recoveries and relative standard deviations of W3 concentrations were It was 86.44% (4.22%), 92.48% (5.84%), and 81.97% (9.32%); the pharmacokinetics of W3 in rats was in line with the one-compartment absorption model. The main pharmacokinetic parameters were t1/2=( 2.99±0.35)h, T(peak)=(2.14±0.27)h. Conclusion: HPLC method for determination of blood concentration of effective component W3 in Rhizoma noteisang was established. The pharmacokinetic parameters of Rhizoma Notoginseng extract were elucidated. The specificity of the method was strong and the result was reliable.
