以黑芝麻无菌苗的下胚轴、幼根、子叶为外植体,以MS+2,4-D1.5mg/L+ZT0.5mg/L为愈伤组织诱导培养基;MS+6-BA2mg/L-IAA0.5mg/L为分化培养基;1/2MS+0.5mg/LNAA+IAA0.25mg/L为生根培养基进行培养。结果表明,幼根能产生少量愈伤组织,尚未分化出苗;子叶愈伤组织能分化出苗,但分化率低;下胚轴愈伤组织不仅可分化出苗(分化率为36%),而且可形成大量具有分化潜力的胚性愈伤组织,分化绿苗的频率可达67%。
The hypocotyls, young roots and cotyledons of sterile black sesame seedlings were used as explants. MS + 2,4-D1.5mg / L + ZT0.5mg / L was used as callus induction medium; MS + 6- BA2mg / L-IAA0. 5mg / L for differentiation medium; 1 / 2MS + 0.5mg / LNAA + IAA0.25mg / L for rooting medium for culture. The results showed that young rootlets could produce a small amount of callus and did not differentiate into seedlings. Cotyledon callus could differentiate and emergence, but its differentiation rate was low. The hypocotyls callus could not only differentiate and shoot (differentiation rate was 36%), but also formed A large number of embryogenic callus with differentiation potential, the frequency of differentiation of green seedlings up to 67%.
