目的探讨丹参酮ⅡA对急性肾功能衰竭(AFR)大鼠肾小管上皮细胞及肾功能的作用。方法将64只大鼠分为4组:对照组、模型组、丹参酮低剂量组和丹参酮高剂量组,每组又分为48 h组和72 h组两个亚组。应用肌肉内注射甘油制备大鼠AFR模型。然后腹腔内注射不同浓度的丹参酮ⅡA进行干预。然后在48 h和72 h检测血尿素氮(BUN)、肌酐(Scr)和24 h尿蛋白定量;同时制作组织切片,观察肾组织结构改变,并应用免疫组织化学方法检测肾组织Ki-67的表达。结果丹参酮高剂量72 h组的各项病理损害指标均低于其他各组(P<0.05);丹参酮高剂量48 h及72 h组的Ki-67阳性细胞计数明显高于其他各组(P<0.05)。结论丹参酮ⅡA对急性损伤的肾小管上皮细胞具有减轻病变和促进再生的作用,可明显改善ARF大鼠的肾功能。
Objective To investigate the effect of tanshinone ⅡA on renal tubular epithelial cells and renal function in rats with acute renal failure (AFR). Methods Sixty-four rats were divided into four groups: control group, model group, low dose tanshinone group and high dose tanshinone group. Each group was divided into two subgroups: 48 h group and 72 h group. Rat AFR model was prepared by intramuscular injection of glycerol. Then intraperitoneal injection of different concentrations of tanshinone Ⅱ A intervention. At 48 h and 72 h, blood urea nitrogen (BUN), creatinine (Scr) and 24 h urinary protein were measured. Tissue sections were made and the changes of renal structure were observed. Immunohistochemistry was used to detect the expression of Ki-67 expression. Results Tanshinone high-dose 72 h group showed lower pathological indexes than other groups (P <0.05). Ki-67 positive cells at high dosage of tanshinone at 48 h and 72 h were significantly higher than those in other groups (P < 0.05). Conclusion Tanshinone ⅡA can relieve the lesion and promote the regeneration of acute renal tubular epithelial cells, and can obviously improve renal function of ARF rats.
